The protein is quite large, and will be the focus of a cross-linking experiment. The goal is to identify the binding partners of our protein of interest, and, if possible, identify the site of cross-linking on our protein. I will be performing mass spectroscopy, including de novo sequencing on selected peptides to determine the sequences of interest. I have not performed tryptic digestion from agarose. The protein is in standard SDS sample buffer. Do any of you have any hints for me before we start? All the best, -laurey Weather inconsequential: Lovely and clear today. My lilacs are in full bloom, and I am eating asparagus from my patch, and morels picked for me on the farm by a cousin----what am I doing in here worrying about mass spectrometry? -- Laurey Steinke, Ph.D Assistant Professor Department of Biochemistry and Molecular Biology Protein Structure Core Facility Mass Spectrometry & Proteomics Facility 985819 Nebraska Medical Center Omaha, NE 68198-5819 T: (402) 559-5176 F: (402) 559-6646 http://www.unmc.edu/pscf/ |
